Objective: Viral nervous necrosis (VNN), as a pandemic disease, infects several kinds of marine fish in the world expect fish of Africa. In expanded mortality of Liza auratus in the Caspian sea in 2003, clinical signs of moribund fish were erratic behavior such as spiral swimming, belly-up and over inflammation of swim bladder. Conformation of VNN in those moribund fish was done by two reference center in Japan and Taiwan by using of RT-PCR and histopathological examinations. Experimental infection in sturgeon was reported. Therefore, transmission of VNN from Liza auratus to sturgeons is possible.
Method & Materials: We examined 87 specimens on brain, eye, sperm and egg from male and female brood stocks that were taken from southern coast of Caspian Sea during spring and summer in 2006 and 2007. In this study, we use IQ2000 RT-PCR kit. RNA extraction was carried out using RNA extraction solution included in the kit based on phenol-chloroform method. RNA qulity was checked by electrophoresis of RNA on 1-2% agarose gel. Target segment of RNA was amplified by RT-PCR and Nested-PCR based on protocol of IQ2000 kit.
Results & Conclusion: Consequences of Nested-PCR were electrophoresed on 2% agarose gel and was stained by Ethidium Bromide. Based on protocol of kit, negative samples indicate a sharp band on 630 bp. In this study all of samples displayed bands with 630 bp, so all of samples were negative.
